Labs that do Nested PCR testing for HHV-6

Wisconsin Viral Research
To our knowledge, this is the only commercial lab in the US that uses nested PCR. They are able to differentiate the A vs. the B variant on samples that are positive. One problem with nested PCR is that there is an increased risk of false positives due to contamination. All laboratories have this problem. Therefore, positive tests should be confirmed with a repeat test or through a procedure called “sequencing the band”. To our knowledge, Wisconsin Viral is also the only lab in the country able to perform immunohistochemical staining on tissue slides to confirm active infections from GI tract biopsy specimens. Order forms and instructions can be downloaded from their web site.

Sample requirements: 1 ml. Frozen plasma or CSF or 1 ADT tube whole blood
Insurance billing: no, but they provide completed forms for insurance billing
Cost, Nested PCR: $275

          IgG & IgM Antibody
-IgG titres at 1:320 or above may mean an active infection)
-normal IgM does not rule out a chronic reactivation
       • Quantitative PCR on serum (can’t detect most low-grade CNS infections)
       • Nested PCR on serum (more sensitive, but still can’t pick up many
         CNS infections)
